Biological Activity I Assay Protocols (From Reference)
Targets
EGTA-AM targets intracellular calcium ions (Ca²⁺). As a membrane-permeable acetoxymethyl ester, it enters cells where it is hydrolyzed by intracellular esterases to release EGTA, a high-affinity Ca²⁺ chelator. EGTA then binds to free Ca²⁺ ions, buffering intracellular calcium levels and modulating calcium-dependent cellular processes.
ln Vitro
Only synchronous excitatory postsynaptic currents (EPSCs) are reduced by EGTA-AM (50 μM) to measure train-generated charge transfer; asynchronous EPSCs are significantly reduced to 58.9 ± 8.1% of control levels [1].
In vitro, EGTA-AM is used as a tool to chelate intracellular calcium ions in cell-based assays. By buffering intracellular Ca²⁺, it allows researchers to study the role of calcium in various cellular functions, including signal transduction, muscle contraction, and neurotransmitter release.
ln Vivo
In vivo activity data for EGTA-AM are limited, as it is primarily used as a research tool for in vitro cellular studies. Its membrane-permeable properties allow it to be used in cell loading experiments, but its application in vivo is limited due to its rapid hydrolysis and metabolism.
Enzyme Assay

For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 25.0 mg/mL clear DMSO stock solution to 400 μL PEG300 and mix evenly; then add 50 μL Tween-80 to the above solution and mix evenly; then add 450 μL normal saline to adjust the volume to 1 mL.
Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH₂ O to obtain a clear solution.
