CSN5i-3 targets CSN5/Jab1, the catalytic subunit of the COP9 signalosome (CSN) complex. The CSN complex regulates the activity of cullin-RING ubiquitin ligases (CRLs) by removing the ubiquitin-like modifier NEDD8 from cullin subunits (deneddylation). By inhibiting CSN5, CSN5i-3 blocks CSN-catalyzed Cul1 deneddylation, leading to accumulation of neddylated cullins and disruption of CRL activity.
