It inhibits CSN-catalyzed Cul1 deneddylation with an IC50 value of 5.8 nM. The compound has the molecular formula C28H29F2N5O2 and molecular weight of 505.56 g/mol. CSN5i-3 has been shown to have cytotoxic effects on various cancer cells and is being studied as a potential anticancer agent.
Biological Activity I Assay Protocols (From Reference)
Targets
CSN5i-3 targets CSN5/Jab1, the catalytic subunit of the COP9 signalosome (CSN) complex. The CSN complex regulates the activity of cullin-RING ubiquitin ligases (CRLs) by removing the ubiquitin-like modifier NEDD8 from cullin subunits (deneddylation). By inhibiting CSN5, CSN5i-3 blocks CSN-catalyzed Cul1 deneddylation, leading to accumulation of neddylated cullins and disruption of CRL activity. This affects the ubiquitination and degradation of various substrates involved in cell cycle regulation and other processes.
ln Vitro
By causing the breakdown of their substrate recognition modules, CSN5i-3 traps CRLs in the neddylated state, which deactivates a portion of CRLs [1].
In vitro, CSN5i-3 has been shown to inhibit CSN-catalyzed Cul1 deneddylation with an IC50 of 5.8 nM. The compound is selective for CSN5 and has been shown to have cytotoxic effects on various cancer cell lines. ln Vivo

For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 25.0 mg/mL clear DMSO stock solution to 400 μL PEG300 and mix evenly; then add 50 μL Tween-80 to the above solution and mix evenly; then add 450 μL normal saline to adjust the volume to 1 mL.
