| Targets |
CNT2 inhibitor-1 targets the concentrative nucleoside transporter 2 (CNT2, also known as SLC28A2). It is a potent inhibitor with an IC50 of 640 nM for the human CNT2 (hCNT2). This compound demonstrates an 81-fold increase in inhibitory activity compared to its parent molecule and a 1500-fold higher potency than other known hCNT2 inhibitors.

|---|---|
| ln Vitro |
CNT2 inhibitor-1 (compound 48) has an inhibitory activity that is 81 times stronger than that of compound 12, the parent molecule. Furthermore, compared to well-known hCNT2 inhibitors such as phloridzin, 7,8,3'-trihydroxyflavone, and 2'-deoxy-5-fluorouridine, CNT2 inhibitor-1 exhibits an inhibitory action that is 1500 times stronger [1]. Although compound 1, or CNT2 inhibitor-1, is an effective inhibitor, its solubility is low [2].
In vitro, CNT2 inhibitor-1 is characterized by its high potency. It achieves an IC50 of 640 nM against hCNT2, demonstrating its effectiveness in blocking nucleoside uptake. Its activity is measured in cellular uptake assays, where it effectively inhibits the transport of radiolabeled or fluorescently labeled nucleosides into cells expressing the CNT2 transporter. |
| ln Vivo |
In vivo, CNT2 inhibitor-1 is used as a pharmacological tool to modulate intracellular nucleoside levels. By inhibiting CNT2, it can affect energy metabolism, nucleoside signaling, and related physiological processes in animal models. Its use helps elucidate the role of CNT2 in various pathophysiological conditions, including cancer and metabolic diseases.
|
| Enzyme Assay |
The in vitro enzyme/receptor binding assay for CNT2 inhibitor-1 is a radioligand uptake assay. Cells overexpressing hCNT2 are incubated with a radiolabeled substrate, such as [3H]-adenosine or [3H]-uridine, in the presence of varying concentrations of the inhibitor. The amount of radioactivity taken up by the cells is measured, and the IC50 is calculated from the inhibition curve.
|
| Cell Assay |
In vitro cell culture studies using CNT2 inhibitor-1 are performed in cells that express the CNT2 transporter. Cells are treated with the inhibitor, and its effect on nucleoside uptake is measured. Additionally, downstream effects such as changes in ATP levels, cell proliferation, or apoptosis can be assessed to understand the functional consequences of CNT2 inhibition.
|
| Animal Protocol |
In vivo animal experiments involve administering CNT2 inhibitor-1 to rodents, typically via intraperitoneal injection. The compound's effects on nucleoside homeostasis are assessed by measuring nucleoside and nucleotide levels in tissues or blood using LC-MS/MS. The compound can also be used to study the role of CNT2 in disease models, such as cancer or ischemia.
|
| ADME/Pharmacokinetics |
CNT2 inhibitor-1 has a molecular weight of 448.47 g/mol and is highly soluble in DMSO. It is typically stored as a powder at -20°C. For in vivo studies, it may be formulated in a suitable vehicle such as a mixture of DMSO, PEG300, and saline. Its stability and pharmacokinetic properties have not been fully characterized, as it is a research compound.
|
| Toxicity/Toxicokinetics |
Toxicological data for CNT2 inhibitor-1 are limited, as it is a research compound. In vitro cytotoxicity assays may be performed in cell lines to assess its safety. However, comprehensive in vivo toxicity studies, including acute and chronic toxicity, have not been reported. It is classified as a research chemical and is not intended for human use.

| Solubility (In Vivo) |
Solubility in Formulation 1: ≥ 2.5 mg/mL (5.57 mM) (saturation unknown) in 10% DMSO + 40% PEG300 + 5% Tween80 + 45% Saline (add these co-solvents sequentially from left to right, and one by one), clear solution.
For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 25.0 mg/mL clear DMSO stock solution to 400 μL PEG300 and mix evenly; then add 50 μL Tween-80 to the above solution and mix evenly; then add 450 μL normal saline to adjust the volume to 1 mL. Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH₂ O to obtain a clear solution. Solubility in Formulation 2: ≥ 2.5 mg/mL (5.57 mM) (saturation unknown) in 10% DMSO + 90% (20% SBE-β-CD in Saline) (add these co-solvents sequentially from left to right, and one by one), clear solution. For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 25.0 mg/mL clear DMSO stock solution to 900 μL of 20% SBE-β-CD physiological saline solution and mix evenly. Preparation of 20% SBE-β-CD in Saline (4°C,1 week): Dissolve 2 g SBE-β-CD in 10 mL saline to obtain a clear solution.  (Please use freshly prepared in vivo formulations for optimal results.) |
