Product Description
Azido-PEG3-acid (N3-PEG3-CH2CH2COOH) is a novel and potent PEG (polyethyleneglycol) analogue used as a PROTAC linker for the synthesis of BI-3663, BI-4216 and BI-0319. As a PEG-based linker, Azido-PEG3-acid is also used as a non-cleavable 3 unit PEG ADC linker for the synthesis of antibody-drug conjugates (ADCs). It is composed of an azido group, PEG spacer, and carboxylic acid. The terminal carboxylic acid can be reacted with primary amine groups in the presence of activators (e.g. EDC, or DCC) forming a stable amide bond. The azide group enables Click Chemistry. The hydrophilic PEG spacer increases solubility in aqueous media.
Azido-PEG3-acid (CAS# 1056024-94-2) is a PEG-based PROTAC linker used in the synthesis of PROTACs. It is also a non-cleavable 3-unit PEG ADC linker used in the synthesis of antibody-drug conjugates (ADCs). It contains an azide group and a terminal carboxylic acid.
Biological Activity I Assay Protocols (From Reference)
Targets
Non-cleavable PEG Linker; PROTAC linker
Azido-PEG3-acid is classified as a PROTAC linker, an ADC linker, and a click chemistry reagent. The azide group can react with alkyne, BCN, or DBCO via click chemistry. The carboxylic acid group can be used for conjugation to primary amines.
ln Vitro
Two distinct ligands, one for the E3 ubiquitin ligase and the other for the target protein, are present in PROTAC and are joined by a linker. PROTAC targets and selectively degrades target proteins by means of the intracellular ubiquitin-proteasome system. An ADC cytotoxin is connected to an antibody by use of an ADC linker to form an ADC.
Azido-PEG3-acid is used as a linker in the synthesis of PROTACs such as BI-3663, BI-4216, and BI-0319. It is also used in ADC synthesis. It is not typically evaluated for direct biological activity as a standalone compound.
ln Vivo
Azido-PEG3-acid is not typically evaluated for in vivo activity as a standalone compound. Its role is in the synthesis of PROTACs or ADCs, which are then evaluated in vivo. The PEG spacer can improve the pharmacokinetic properties of the conjugated molecules.
Enzyme Assay
Azido-PEG3-acid is a chemical reagent, and its purity and identity are confirmed using analytical techniques such as NMR and HPLC. It is not typically used in enzyme or receptor binding assays. Its reactivity can be assessed by reacting it with amines or alkyne-containing compounds.
Cell Assay
Azido-PEG3-acid is not typically used in cell-based assays as a standalone compound. Its primary application is in the synthesis of PROTACs or ADCs, which are then tested in cell-based assays for their activity.
Animal Protocol
Azido-PEG3-acid is not typically used in in vivo animal studies as a standalone compound. It is used to synthesize PROTACs or ADCs, which are then evaluated in animal models. The PEG spacer can influence the pharmacokinetics of the conjugated molecules.
ADME/Pharmacokinetics
Azido-PEG3-acid has a molecular weight of 261.28 and a formula of C9H17N3O5. It contains an azide group and a terminal carboxylic acid. It is a monodisperse PEG molecule that is soluble in aqueous media.
Toxicity/Toxicokinetics
Specific toxicology data for Azido-PEG3-acid are not detailed in the available literature. As a linker used in PROTAC or ADC research, its toxicity is typically assessed as part of the final conjugated product. Standard safety precautions should be taken when handling this compound.
