Immunotherapy for glioma is an attractive strategy yet faces significant barriers: (i) inefficient delivery across the blood-brain barrier (BBB), and (ii) a profoundly immunosuppressive microenvironment. To address these constraints, we engineered a biomimetic nanocarrier, where EGCG/Zn core loaded MSA-2 and siRNA-SLC43A2 was coated with glioma cell membrane-liposome fusions. Incorporation of an Angiopep-2 (Ang-2) targeting peptide was further intended to facilitate BBB transcytosis via LRP1-mediated transport. Leveraging its excellent immune activation capability and targeting specificity, this nanocarrier successfully traversed the BBB and delivered

therapeutic agents precisely into glioma cells. MSA-2 activated STING pathway to trigger innate immune response and induce the CD8⁺ T cells recruiting. Complementarily, siRNA targeting SLC43A2 reversed methionine depletion and alleviated CD8⁺ T cell exhaustion and apoptosis by downregulating the methionine transporter in tumor cells. These two mechanisms synergistically enhanced anti-glioma immunotherapy by initiating the anti-tumor immune response and augmenting its therapeutic efficacy, respectively. This synergistic immunotherapy provides a promising novel strategy to overcome the challenges of glioma immunotherapy
