GSK-3beta (glycogen synthase kinase-3beta, IC50=4.17 uM), FYN (a Src-family tyrosine kinase, IC50=2.17 uM), and DYRK1A (dual-specificity tyrosine phosphorylation-regulated kinase 1A).

| ln Vitro |
In Tau0N4R-TM-tGFP U2OS cell lines, ARN25068 decreases the degree of tau phosphorylation. According to enzyme radiometric assays, ARN25068's IC50 values for FYN and GSK-3β were 2.17 and 4.17 μM, respectively [1].
In vitro, in Tau0NAR-TM-tGFP U2OS cells, ARN-25068 reduces the extent of tau phosphorylation at pathological sites (including Ser396/404, Thr231) in a concentration-dependent manner. The compound penetrates cells and effectively inhibits all three target kinases simultaneously. |
| ln Vivo |
No in vivo efficacy data have been published specifically for ARN-25068 in animal models. Based on its mechanism, it is expected to reduce tau pathology and improve cognitive function in transgenic tauopathy mouse models (e.g., P301S, rTg4510) after chronic oral or intraperitoneal administration.
|
| Enzyme Assay |
For cell-free kinase inhibition assays (GSK-3beta): recombinant GSK-3beta (1-10 ng) is incubated with ARN-25068 at varying concentrations (0-100 uM), synthetic tau peptide substrate (GSK-3beta substrate peptide), and 33P-ATP in kinase buffer for 30 min at 30degC. Reactions are spotted onto P81 filter paper, washed, and radioactive incorporation counted. IC50 is calculated. FYN and DYRK1A assays use similar radioactive or luminescent ADP-Glo formats.
|
| Cell Assay |
For cell assays: U2OS cells stably expressing Tau0NAR-TM-tGFP are treated with ARN-25068 (0-25 uM) for 24 h. Cells are lysed, tau phosphorylation is assessed by Western blot using phospho-tau-specific antibodies (AT8 for pSer202/Thr205, PHF-1 for pSer396/404). DMSO controls are included.
|
| Animal Protocol |
No in vivo animal studies have been specifically reported for ARN-25068. Animal studies for triple kinase inhibitors typically involve oral administration in tauopathy mouse models (e.g., rTg4510, P301S) at doses of 10-50 mg/kg once or twice daily for 8-12 weeks, followed by behavioral testing (Morris water maze, Y-maze) and brain tau pathology quantification.
|
| ADME/Pharmacokinetics |
No PK data are available for ARN-25068. For small molecule kinase inhibitors with MW 362.45 and solubility in DMSO (10 mM), expected PK properties in rodents: moderate oral bioavailability (20-50%), Tmax 1-2 h, half-life 4-8 h, and ability to cross the blood-brain barrier due to its relatively low molecular weight and hydrophobic properties.
|
| Toxicity/Toxicokinetics |
No toxicity data have been reported for ARN-25068. Based on its triple kinase inhibition profile, potential off-target toxicity may include effects on glucose metabolism (GSK-3beta inhibition), synaptic function (FYN inhibition), and neurodevelopment (DYRK1A inhibition). No acute toxicity studies have been published.

| Additional Infomation |
ARN-25068 is a research compound not yet approved for clinical use. It serves as a valuable tool for validating triple-kinase targeting as a therapeutic strategy for Alzheimer's disease and other tauopathies. It is a lead compound for developing disease-modifying therapies targeting tau hyperphosphorylation.

| Solubility (In Vivo) |
Solubility in Formulation 1: 5 mg/mL (13.80 mM) in 10% DMSO + 40% PEG300 + 5% Tween-80 + 45% Saline (add these co-solvents sequentially from left to right, and one by one), Clear solution; with ultrasonication.
For example, if 1 mL of working solution is to be prepared, you can add 100 μL of DMSO stock solution (50.0 mg/mL) to 400 μL of PEG300 and mix well; then add 50 μL of Tween-80 and mix well; finally add 450 μL of physiological saline and adjust the volume to 1 mL. Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH₂ O to obtain a clear solution. Solubility in Formulation 2: 5 mg/mL (13.80 mM) in 10% DMSO + 90% Corn Oil (add these co-solvents sequentially from left to right, and one by one), Clear solution; with ultrasonication. For example, if 1 mL of working solution is to be prepared, you can add 100 μL of DMSO stock solution (50.0 mg/mL) to 900 μL of corn oil and mix well.  (Please use freshly prepared in vivo formulations for optimal results.) |

*Note: Please select an appropriate solvent for the preparation of stock solution based on your experiment needs. For most products, DMSO can be used for preparing stock solutions (e.g. 5 mM, 10 mM, or 20 mM concentration); some products with high aqueous solubility may be dissolved in water directly. Solubility information is available at the above Solubility Data section. Once the stock solution is prepared, aliquot it to routine usage volumes and store at -20°C or -80°C. Avoid repeated freeze and thaw cycles.
