Pentyl-m-cresol is a phenolic compound with a structure similar to m-cresol, but with an pentyl group substituted at the 6-position. It is an antibacterial agent with similar functions to m-cresol. Pentyl-m-cresol is an over-the-counter antibacterial agent available in Canada in various lozenges for the treatment of sore throat and minor oral infections. Pentyl-m-cresol is often formulated with dichlorobenzyl alcohol and menthol in common sore throat lozenges, such as Strepsils. Acute sore throat (pharyngitis) is one of the most common conditions seen in children at primary care facilities. Pharyngitis is usually caused by a virus, is generally benign, and self-limiting. Clinically proven over-the-counter throat lozenges provide rapid and effective relief of acute sore throat symptoms and are playing an increasingly important role in the treatment of this condition. Indications: Sore throat, mild oral and throat infections… Mechanism of Action: Its antiviral mechanism is not fully understood, but studies suggest it may work by denaturing viral external protein spikes, pH-induced rearrangement of attachment proteins' tertiary structure, or selectively acting on the viral lipid membrane/protein-lipid interactions. Pentocresol is an antibacterial and antiviral drug that blocks voltage-gated sodium channels in a manner similar to local anesthetics. Pharmacodynamics: Pentocresol throat lozenges significantly reduce the infectivity of certain infectious viruses in the throat and cough droplets, thereby reducing the chance of human-to-human transmission. It also relieves sore throat/throat irritation symptoms. Pentocresol is believed to inhibit inflammatory and pain mediators involved in oral and throat mucosal inflammation and sore throat.

Solubility in Formulation 1: ≥ 2.5 mg/mL (14.02 mM) (saturation unknown) in 10% DMSO + 40% PEG300 + 5% Tween80 + 45% Saline (add these co-solvents sequentially from left to right, and one by one), clear solution.
For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 25.0 mg/mL clear DMSO stock solution to 400 μL PEG300 and mix evenly; then add 50 μL Tween-80 to the above solution and mix evenly; then add 450 μL normal saline to adjust the volume to 1 mL. Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH₂ O to obtain a clear solution. Solubility in Formulation 2: 2.5 mg/mL (14.02 mM) in 10% DMSO + 90% (20% SBE-β-CD in Saline) (add these co-solvents sequentially from left to right, and one by one), clear solution; with ultrasonication. For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 25.0 mg/mL clear DMSO stock solution to 900 μL of 20% SBE-β-CD physiological saline solution and mix evenly.
