Afoxolaner is an orally bioactive isoxazoline insecticide/miticide against scapula lizards. Afoxolaner works on the γ-aminobutyric acid receptors (GABA) and glutamate receptors of insects, inhibiting the absorption of chloride ions regulated by GABA and glutamate, resulting in excessive neuronal stimulation and joint damage. Animals die.

ln Vivo
Afoxolaner's minimum effective dose of 2.5 mg/kg, when taken as a single oral therapy, was found to be 98% to 100% effective on day 30 and to be effective in eliminating pre-existing scapularis tick infections and weekly reinfections up until day 23 [1].

Drug Indication
Treatment of flea (Ctenocephalides felis and C. canis) infestations. The product can be used as part of a treatment strategy for the control of flea allergy dermatitis (FAD). , , Treatment of tick (Dermacentor reticulatus, Ixodes ricinus, Rhipicephalus sanguineus) infestations. , , Treatment of demodicosis (caused by Demodex canis). , , Treatment of sarcoptic mange (caused by Sarcoptes scabiei var. canis). ,
Treatment of flea infestation in dogs (Ctenocephalides felis and C. canis)  for at least 5 weeks. The product can be used as part of a treatment strategy for the control of flea allergy dermatitis (FAD). Treatment of tick infestation in dogs (Dermacentor reticulatus, Ixodes ricinus, Ixodes hexagonus, Rhipicephalus sanguineus). One treatment kills ticks for up to one month. Fleas and ticks must attach to the host and commence feeding in order to be exposed to the active substance.  Treatment of demodicosis (caused by Demodex canis). Treatment of sarcoptic mange (caused by Sarcoptes scabiei var. canis).

Solubility in Formulation 1: ≥ 2.25 mg/mL (3.59 mM) (saturation unknown) in 10% DMSO + 40% PEG300 + 5% Tween80 + 45% Saline (add these co-solvents sequentially from left to right, and one by one), clear solution.
For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 22.5 mg/mL clear DMSO stock solution to 400 μL PEG300 and mix evenly; then add 50 μL Tween-80 to the above solution and mix evenly; then add 450 μL normal saline to adjust the volume to 1 mL.
Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH₂ O to obtain a clear solution.

Solubility in Formulation 2: ≥ 2.25 mg/mL (3.59 mM) (saturation unknown) in 10% DMSO + 90% Corn Oil (add these co-solvents sequentially from left to right, and one by one), clear solution.
For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 22.5 mg/mL clear DMSO stock solution to 900 μL of corn oil and mix evenly.
