Biological Activity I Assay Protocols (From Reference)
Targets
7-Methylguanine primarily targets PARP1 (Poly(ADP-ribose) polymerase 1), an enzyme involved in DNA repair. It acts as a competitive inhibitor of PARP1 with a Ki of 61 μM, competing with the substrate NAD+. By inhibiting PARP1, the compound interferes with DNA repair mechanisms, leading to anticancer activity. As a natural nucleic acid metabolite, it serves as a probe for protein-DNA interactions. Its anticancer activity has been demonstrated against uterine sarcoma and colon adenocarcinoma.
ln Vitro
In vitro, 7-Methylguanine (0-200 μM) inhibits the activity of PARP1 enzyme in a dose-dependent manner. It is a competitive PARP-1 inhibitor with an inhibition constant (Ki) of 61 μM. 7-Methylguanine (0-450 μM) suppresses the catalytic activity of PARP-1, thereby preventing the dissociation of the PARP-1-DNA complex. After 24 hours of treatment with 7-Methylguanine (0.02-0.2 mg/mL), no DNA damage was observed in immortalized human renal epithelial cells (NKE-hTERT). The compound exhibits anticancer activity against uterine sarcoma and colon adenocarcinoma.
ln Vivo
In vivo, in CBA, BALB/c, and C57BL/6 mice, 7-Methylguanine (50 mg/kg, administered orally, three times per week for 4 weeks) did not cause adverse reactions or morphological changes, while it inhibited tumor growth in CBA mice with uterine sarcoma US-322. In BALB/c mice transplanted with Akatol tumors, 7-Methylguanine (50 mg/kg, administered via gavage, three times per week) effectively penetrated tumor tissues and suppressed the growth of colon adenocarcinoma. The compound is orally available.
Enzyme Assay
In vitro enzyme assays for 7-Methylguanine involve measuring PARP1 activity inhibition. PARP1 activity is assessed by measuring the incorporation of radiolabeled NAD+ into poly(ADP-ribose) polymers. The compound is added at varying concentrations (0-200 μM), and IC50 or Ki values are calculated from dose-response curves. Competitive inhibition is confirmed by varying substrate (NAD+) concentrations. DNA repair assays may be performed to assess functional consequences of PARP1 inhibition. Animal Protocol
In vivo animal studies for 7-Methylguanine are conducted in mouse models of uterine sarcoma and colon adenocarcinoma. CBA mice with uterine sarcoma US-322 or BALB/c mice transplanted with Akatol tumors are treated with 7-Methylguanine at 50 mg/kg, administered orally or via gavage, three times per week for 4 weeks. Tumor growth is monitored by caliper measurements. Body weight and clinical signs are monitored to assess toxicity. Tumor tissues are collected for histopathological and biomarker analysis. PARP1 inhibition in tumors is confirmed by measuring PAR polymer levels. Studies are conducted in accordance with institutional animal care guidelines.
ADME/Pharmacokinetics
7-Methylguanine (MW 165.15 g/mol, C6H7N5O) is a small molecule PARP1 inhibitor. It is soluble in 1M NaOH (80 mg/mL, 484.41 mM) and water (3.3 mg/mL, 19.98 mM, when pH is adjusted to 11 with 1M NaOH). The compound is stable as a powder at -20°C for up to 3 years and in solvent at -80°C for 1 year. It is orally available. Pharmacokinetic parameters such as half-life, bioavailability, and tissue distribution have been characterized in preclinical studies.
Toxicity/Toxicokinetics
7-Methylguanine is generally well-tolerated in preclinical studies. In CBA, BALB/c, and C57BL/6 mice, 7-Methylguanine (50 mg/kg, administered orally, three times per week for 4 weeks) did not cause adverse reactions or morphological changes. The compound is a natural nucleic acid metabolite with a favorable safety profile. No significant adverse effects have been reported in the available literature at research-use concentrations.
