TP-α (10 μM; incubated on ice for 30 min) can bind directly and selectively to glucagon, with a dissociation constant (Kd) of 65 μM, and its fluorescence intensity increases 9-fold at a glucagon concentration of 1 mg/mL [1]. TP-α (0.5-1 μM; incubated for 1 h) can selectively label glucagon-secreting α TC1 cells, while the fluorescence intensity on β TC6 cells and acinar 266-6 cells is negligible [1]. TP-α (1-2 μM; incubated for 1 h or 24 h) has extremely low cytotoxicity to α TC1 cells and β TC6 cells [1]. TP-α (single photon concentration of 1 μM; two photon concentration of 10 μM) exhibits single photon absorption/emission at 370/510 nm (quantum yield of 0.69), two photon interaction cross section of 107 GM, optimal two photon excitation wavelength of 750 nm, and emission wavelength of 430-530 nm [1].
