NC762 (1–30 mg/kg, intraperitoneal injection, once every 7 days for a total of 4 times) dose-dependently reduced tumor volume in NSG mice subcutaneously inoculated with 1E06 624Mel.hB7H4 cells and 3E05 human peripheral blood mononuclear cells (PBMCs) [2]. NC762 (10 mg/kg, intraperitoneal injection, once every 4 days for a total of 7 times) inhibited anti-B7H4-mediated, NK cell-involved tumor growth and prolonged the survival of NSG mouse models subcutaneously inoculated with a mixture of 1E06 624Mel.hB7H4 cells and 1E06 total PBMCs or NK cell-depleted PBMCs [2]. NC762 (10 mg/kg, intraperitoneal injection, once every 7 days for a total of 4 times) inhibited tumor growth in NSG mouse models subcutaneously inoculated with 1E06 624Mel.hB7H4 cells, regardless of the presence of PBMCs [2]. The tumor growth inhibition activity of NC762 (10 mg/kg, intraperitoneal injection, once every 7 days for a total of 4 times) is similar to that of the low Fc affinity mutant of NC762, suggesting that its tumor growth inhibition mechanism may not depend on ADCC[2].
