Antibodies to bovine type I and type III collagen and their precursor form procollagen were produced in rabbits and rendered specific for the immunizing antigen by immunoadsorption. These purified antibodies showed distinct immunofluorescence staining on frozen sections of both bovine and human connective tissue at concentrations as low as 1-10 mug/ml. Antibodies to type III collagen and procollagen reacted with reticulin in liver and spleen, with fascicles around tendons and with the upper portion of the dermis. Antibodies to type I collagen and procollagen reacted with skin and fiber bundles in tendon but did not stain reticulin. No reaction was observed with cartilage collagen or with kidney glomerular basement membrane. [1]

Collagen is the most abundant component of the extracellular matrix. Elastin works with collagen to give the lungs, aorta and skin elastic recoil. These fibers are highly resistant to proteolysis, but a few matrix metalloproteinases (MMPs) can break them down. Currently, we have a good understanding of how MMPs move to the susceptible sites of collagen and untangle the triple helix structure of collagen monomers. Research focuses on the key role of the heme-binding protein-like domain of interstitial collagenase or the collagen-binding domain of gelatinase in untangling the triple helix structure of collagen. The collagen-binding domains of MMP-2 and MMP-9 also promote the breakdown of elastin, while the distal extra-tertiary site of the catalytic domain of MMP-12 promotes the breakdown of elastin. [2]
