PARG

Poly (ADP-ribose) glycohydrolase (PARG) is the primary hydrolase involved in the degradation of poly(ADP-ribose) (PAR).Both endo- and exo-glycohydrolase activity are present in PARG, with the latter activity being preferentially carried out by binding to the two ADP-ribose residues that are furthest from the PAR chain. Free PAR and mono ADP-ribose moieties are produced by these various catalytic methods, respectively. By ADP-ribose pyrophosphohydrolases like the NUDIX family, the free mono ADP-ribose is then converted into AMP and ribose 5′ phosphate. While ribose 5′ phosphate is a precursor to many biomolecules including DNA, RNA, and ATP, AMP is used in the ATP reformation process as well as various metabolic and cell signaling pathways. The production of free PAR chains as a result of hyper-PARP activation is thought to be the main cause of endo-glycohydrolase activity, which is then associated with apoptosis and thought to function as a death signal.
