Transforming growth factor beta (TGF-β) is a multifunctional cytokine of the TGF superfamily which mainly includes 3 mammalian isoforms (TGF-β 1 to 3, HGNC symbols TGFB1, TGFB2, TGFB3) and many other signaling proteins. Abnormalities of the TGF-β receptors and Smads have been found in various tumors such as colorectal cancers and pancreatic cancers. In addition, TGF-β/BMP signaling is also involved in osteoblast differentiation, chondrocyte differentiation, skeletal development, cartilage formation, bone formation, bone homeostasis, and related human bone diseases caused by the disruption of TGF-β/BMP signaling.
