Atezolizumab (also known as MPDL3280A; trade name: Tecentriq) is a novel and selective humanized monoclonal IgG1 antibody against programmed death ligand 1 (PD-L1). In May 2016 FDA granted accelerated approval to atezolizumab for locally advanced or metastatic urothelial carcinoma treatment after failure of cisplatin-based chemotherapy.[3] The confirmatory trial (to convert the accelerated approval into a full approval) failed to achieve its primary endpoint of overall survival. In October 2016, FDA approved atezolizumab for the treatment of patients with metastatic non-small cell lung cancer (NSCLC) whose disease progressed during or following platinum-containing chemotherapy. Patients with EGFR or ALK genomic tumor aberrations should have disease progression on FDA-approved therapy for these aberrations prior to receiving atezolizumab.

Osteosarcoma cells are treated with atezolizumab (0, 2.5, 5, 10, 20, and 40 µg/ml; 24 hours; HOS and 143B cells) to induce immune-dependent apoptosis and reduce proliferation [1]. Because of its effects on mitochondrial function, atezolizumab leads to an imbalance between antioxidants and oxidants. By triggering the JNK pathway, atezolizumab causes osteosarcoma cells to undergo mitochondria-related apoptosis. In osteosarcoma cells, atezolizumab (10 µg/ml; 24 hours; HOS and 143B cells) activates autophagy [1].

Injection Formulation 1: DMSO : Tween 80: Saline = 10 : 5 : 85 (i.e. 100 μL DMSO stock solution → 50 μL Tween 80 → 850 μL Saline)(e.g. IP/IV/IM/SC) *Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH ₂ O to obtain a clear solution.
